ANC1 was identified in the primary haploid screen as CHI with act1?. It is a subunit (30 kDa) of the TFIID, TFIIF, and SWI/SNF complexes, is involved in RNA polymerase II transcription initiation and in chromatin modification, and contains a YEATS domain. Anc1p is also known as TafII30. It is part of the Ino80 complex which includes: Act1, Arp4, Arp5, Arp8, Ies1, Ies3, Ino80, Nhp10, Rvb1, Rvb2, and Taf14. Anc1p also immunoprecipitates with Rpb9, Taf2, Sap185, Mus81, Taf14, Rpb5, Rpo21, and Tfg1 and is in a purified complex with Snf5. The anc1? null shows reduced fitness in YPD and is reported to have a depolarized actin cytoskeleton (see below). Anc1p-GFP localizes exclusively to the nucleus with 3120 copies per cell as per the O'Shea GFP web site. ANC1 has no paralog but has some homology to Sas5p which is a histone acetylase that regulates transcription. Actin staining of the null haploid showed very large cells and a high percentage of cells with gross morphology defects, extremely numerous but thin cables-scored as having strong defects
